Job Description
Missions
The engineer will work to characterize the function of perception and degradation pathways of hormonal signals (auxin, cytokinin) involved in the development of plant architecture in the model moss Physcomitrium patens.
Activity
• The engineer will develop and use methods of molecular biology and molecular genetics (cloning, plant genetic transformation, genotyping) to study the actors of perception and degradation pathways of hormonal signals and their targets.
• The engineer will develop and use plant phenotyping methods (morphological analysis of meristems, branch quantification, growth monitoring under different hormonal conditions).
• The engineer will develop and use quantitative imaging methods on living tissues with temporal follow-up (time course) in planta, with a particular focus on moss meristems.
• The engineer will implement protocols for in planta hormone quantification.
• The engineer will develop protocols and implement bulk and single-cell RNA-seq experiments, and associated bioinformatic analyses, as well as Hybridization Chain Reaction (HCR) experiments for the spatial anchoring of transcriptomic data.
• He/she will use standard and/or laboratory-developed software (R, Python, Seurat, Imaris, FIJI) for image analysis and bulk/scRNA-seq data analysis.
• He/she will participate in the design of large-scale mutagenesis experiments (CRISPR/Cas9, UV) and implement them autonomously.
• He/she will contribute to the writing of scientific publications and reports.
Your Profil
Skills
• Excellent mastery of plant development biology concepts;
• Solid experience in biology and molecular genetics;
• Solid experience in in vitro plant culture;
• Good mastery of photonic imaging (transmission, confocal) and image analysis;
• Good mastery of computational tools for RNA-seq data analysis;
• Autonomy in the design, execution, and analysis of experimental results;
• Excellent ability to integrate and work in a team;
• Excellent ability to present results in writing and orally;
• Proficiency in English: reading, writing, speaking.
Your Work Environment
The engineer will work within the SIGNAL team ("Signaling and Developmental Dynamics") at the Plant Reproduction and Development Laboratory (RDP, CNRS UMR 5667, Lyon).
