Job Description
Missions
Cellular plasticity is a major mechanism by which tumor cells adapt to microenvironmental constraints, particularly nutrient deprivation. In lymphomas, resistance to chemotherapy, including asparaginase-based treatments, is partly driven by the activation of molecular pathways that enable tumor cells to adapt to stress and maintain their survival. This project aims to characterize the signaling pathways and RNA Damage Response (RDR) mechanisms involved in cellular plasticity and therapeutic escape in lymphoma. In particular, we will investigate how alterations in RNA integrity and function are sensed and integrated by tumor cells to remodel their cellular state, metabolism, and survival capacity in response to treatment. The overall objective is to identify the molecular mechanisms linking cellular stress, plasticity, and therapeutic resistance, thereby uncovering new cellular and metabolic vulnerabilities that could be exploited to develop targeted therapeutic strategies in oncology.
Activity
With a PhD in cellular and molecular biology, the successful candidate should have a strong scientific background, both theoretical and practical, in cellular stress signaling pathways, regulation of ribosome biogenesis, translation, and cell fate, as demonstrated by scientific publications.
The successful candidate is expected to demonstrate a high degree of autonomy, rigor, excellent organizational skills, and strong scientific communication and reporting abilities. They will also contribute to the collective activities of the research team and laboratory, as well as to the dissemination and valorization of research findings.
Your Profil
Skills
* Cell culture and lymphoma models: analysis of cellular responses, adaptation, and cell fate following exposure to different chemotherapeutic agents, using liquid culture lymphoma models (cell lines and/or primary samples). Application of RNA interference, CRISPR/Cas9, and pharmacological approaches.
* Biochemistry and molecular biology: characterization of cellular stress signaling pathways and treatment-induced alterations in translation, using Western blotting, immunoprecipitation (IP), immunofluorescence, polysome profiling, RNA analyses, and proteomics.
* Flow cytometry (FACS): analysis of cellular phenotypes, proliferation, viability, and cell death.
* Omics data analysis: experience in the processing and analysis of omics datasets using R would be an advantage, particularly for transcriptomics, proteomics, and Ribo-seq data.
* Scientific communication and research valorization: independent analysis and interpretation of experimental data, rigorous maintenance of an electronic laboratory notebook, contribution to the preparation of grant applications and scientific manuscripts, and presentation of research findings at team meetings and scientific conferences.
Your Work Environment
This position is based at the Laboratoire de Biologie Tissulaire et Ingénierie Thérapeutique (LBTI – CNRS UMR 5305, Université Claude Bernard Lyon 1), located in the heart of the dynamic Lyon-Gerland Biodistrict. The successful candidate will join the research team led by Dr Cédric Chaveroux and contribute to the development of an INCa PLBIO-funded project, conducted in close collaboration with academic partners in Nice and Paris. Occasional travel as part of these collaborations will be required.
