Missions

Context: Analyzing the neuro-immune mechanisms which result from persistent brain infection that may alter neuronal DNA damage response and their contribution to the progression of cognitive decline in Alzheimer's disease.

This project is part of the Occitania Region-sponsored INSPIRE initiative to shed light on mechanisms involved in aging, and to identify strategies to promote healthy aging. This project consists in analyzing the mechanisms underlying cognitive decline in Alzheimer's disease (AD), using murine models of the disease. In this neurodegenerative and neuroinflammatory disease, we will explore the impact of the inflammatory response on the severity and the progression of AD-related symptoms. Moreover, we plan to analyze the consequences of infections by neurotropic pathogens, such as the parasite Toxoplasma gondii, on the evolution of AD. We found that several proinflammatory cytokines and neuronal infection by Toxoplasma gondii cause DNA double strand breaks to accumulate into the DNA of neurons. Finally, we will decipher what role these various processes may play in epigenetic.

Activities

Our project aims to: (i) study the impact of latent toxoplasmosis on the severity and progression of AD-related pathology in an inducible AD mouse model and on immune phenotypes and determine the status of senescence of immune cells in relation with parameters of their DSB response. (ii) determine how the resident memory T cell response contribute in the long-term to cognitive decline and aggravation of AD-related behavioral deficits. (iii) modulate anti-parasite immune response, to decipher their role in the severity of AD. This project will be run in parallel with a study on a human cohort. By shedding light on new correlates between peripheral and central immune profiles, brain infections and cognitive decline in AD, our project aims at building knowledge on the cellular mechanisms involved in AD and at identifying new biomarkers that may be predictive of the evolution of these pathology.

Work Context

As a post-doctoral fellow, you will be supervised by Elsa Suberbielle and Nicolas Blanchard. Our teams are part of the Center of Pathophysiology Toulouse Purpan (CPTP), a research institute co-affiliated with INSERM, CNRS and University of Toulouse. As such, the CPTP is a stimulating and cross-disciplinary environment for young researcher to build a career in immunology, infectious diseases in the context of neuroinflammatory of neurodegenerative diseases. Our cutting-edge core facilities offer their on-site expertise. Our environment is multicultural thanks to the hosting of students and post-docs of all kinds of citizenship. Weekly lab meetings and seminars are held in English. The CPTP is located in Toulouse, an attractive city with fast economic growth, dynamism of its high technologies, and job opportunities in Science (and a better weather than in Paris).
This Postdoctoral position is part of a 24-month long funding from the INSPIRE project in Toulouse, with the possibility of a one-year prolongation thanks to additional funding. Depending on prior experience and employment, the salary ranges between 2036 and 2337 € (net income) per month, including health-care benefits. The Start date has to be prior the 1st of July 2020.

Constraints and risks

Working with rodents, work in confined environment, flexible research schedules requiring occasional completion of tasks after business hours and/or during weekends, missions in other laboratories in France or abroad.
