Job Description
Missions
-The successful candidate will be recruited as part of the ANR project "PROTECT-DN". Their responsibilities will include:
Characterizing the excitability of murine sensory neurons in a model of type 2 diabetic neuropathic pain (induced by a high-fat, high-sugar diet) compared to control animals, by comparing genetic inhibition models of the Cav3.2 calcium channel. Methodological approaches will include ex vivo electrophysiology of sensory fibers and neurons (skin-nerve preparations and patch-clamp recordings), as well as in vivo calcium imaging or extracellular electrophysiology in anesthetized mice (miniscope videofluorimetry or single/multi-unit action potential recordings).
Characterizing the excitability of murine sensory neurons in the same type 2 diabetic neuropathic pain model, comparing animals chronically treated with a T-type calcium channel pharmacological inhibitor currently in clinical development. The methodological approaches will be identical to those outlined in Point 1.
Conducting longitudinal tracking of mechanical and thermal pain sensitivity thresholds in cohorts of mice subjected to the type 2 diabetes model, utilizing a novel automated behavioral analysis platform (RAMalgo system) ensuring unbiased assessment. A development phase will consist of coupling this automated pain evaluation with electrophysiological or videofluorimetric recordings.
Participating in the development of a genetic approach aimed at inducing targeted and progressive degeneration of the sensory neurons most vulnerable to type 2 diabetic neuropathy, and phenotypically characterizing this mouse model in vitro and in vivo.
This work contributes to the validation of a preventive therapeutic strategy for diabetic neuropathic pain, opening concrete perspectives for translating preclinical research into clinical applications.
(12-month contract, renewable)
Activity
Characterize the excitability of peripheral sensory fibers and their modulation by type 2 diabetic pathology (in the high-fat, high-sugar diet model) using ex vivo electrophysiological approaches (skin-nerve technique), in vivo action potential recordings, and videofluorimetry.
Implement an innovative automated platform for behavioral analysis of somatosensation in mice, incorporating cutting-edge technical developments, notably co-recording of electrophysiological signals.
Manage and execute the breeding of transgenic mouse lines required for project needs.
Analyze, interpret, and disseminate experimental data through scientific publications in peer-reviewed international journals.
Your Profil
Skills
Electrophysiological Implementation and Analysis: Mastery of electrophysiological recordings in patch-clamp, skin-nerve preparations, and in vivo single/multi-unit activity for studying cellular excitability, along with data analysis and processing.
Animal Experimentation: Expertise in in vivo animal experimentation within the field of pain and/or other neuroscience domains (neurodegenerative diseases, neurological disorders, metabolic syndrome, etc.). Project Designer certification (Level 1) and surgical competence are mandatory.
Desirable Skills: Proficiency in miniscope imaging, fiber photometry, or in vivo electrophysiology would be a strong asset for the project.
Personal and Interpersonal Skills: Strong ability to perform literature reviews and synthesize findings, solid background in neuroscience and electrophysiology, effective scientific communication skills, autonomy, and proven ability to work as part of a team.
Your Work Environment
The project will be conducted at the Institute of Functional Genomics (IGF) within a research team internationally recognized for its work on T-type calcium channels in somatosensation, chronic pain, and pleasant touch. The recruited researcher will benefit from the multidisciplinary expertise of the team and the Institute, as well as the specialized capabilities of project partners at an institute dedicated to diabetes research, alongside collaborators at UT Dallas (USA).
The IGF is located on the Arnaud de Villeneuve campus, an active participant in the "Objectif Employeur Pro-Vélo" program.
