Missions
The postdoctoral project is part of the SymbiOcean project funded by an ERC consolidator grant, which aims to study plankton photosymbiosis from the ocean at the subcellular, molecular and ecological scales. The main objective of the postdoctoral project is to understand the morphological changes of a symbiotic marine alga by applying 3D cryo-electron microscopy (cryo-TEM), with a particular focus on the strcutral organization of the chloroplast in free-living versus symbiotic stage. The researcher's mission will be to collect and cryofix planktonic symbioses in the ocean during the TREC expedition (https://www.embl.org/about/info/trec/expedition/) for observation using 3D electron microscopy. The researcher will benefit from the team's network of collaborations in Grenoble and within the TREC consortium.
Activités
- Sampling planktonic symbioses in the field
- Fluorescence microscopy in cryogenic mode
- Image analysis of tomograms (segmentation, 3D reconstruction) from 3D electron microscopy
- Communication through international meetings and workshops.
Compétences
- Cell biology or biophysics using electron and/or correlative microscopy.
- Knowledge and expertise in sample preparation for 3D electron microscopy (FIB-SEM, cryo-TEM) and fluorescence microscopy in cryo mode.
- OR Knowledge and expertise in image analysis for 3D reconstruction after FIB-SEM or cryo-TEM.
- Ability to work independently in an interdisciplinary and international environment, and to interact in a young and dynamic research group.
Contexte de travail
The researcher will be part of Photosymbiosis team (photosymbiosis.com) of the Cellular and Plant Physiology Physiology Laboratory (UMR 5168 - CNRS -CEA - Univ. Grenoble Alpes - INRAe) in Grenoble. The team's projects aim to better understand the functioning of unicellular photosymbiosis by combining sub-cellular microscopy approaches (nanoSIMS, Synchrotron X-Ray fluorescence, 3D electron microscopy). Locally, the project will work in close collaboration with the IRIG institute: INAC platform (IRIG) and the Institut de Biologie Structurale (IBS, Grenoble). This position is part of an ERC consolidator-funded project (SymbiOCEAN) led by the Photosymbiosis team. Numerous interactions will take place with imaging platforms, and local and international local and international partners during meetings and field sampling.
