Job Description
Missions
The successful candidate will work on the development and validation of new techniques for the design and production of synthetic human mitochondrial DNA. They will develop various approaches aimed at modularising the design of synthetic DNA in order to explore the biological consequences of the sequence rearrangements observed in patients with mitochondrial disorders. The successful candidate will be responsible for the production of synthetic mitochondrial DNA within the MIDAS consortium.
Activity
Main responsibilities
• Develop new methods for the production of synthetic mitochondrial DNA.
• Explore in vitro and in vivo assembly techniques.
• Establish various protocols for large-scale production whilst preserving sequence integrity.
• Contribute to the analysis, interpretation, traceability and utilisation of the data obtained.
Secondary activities
• Attend project meetings, team meetings, seminars and presentations of results.
• Liaise closely with project partners and other members of the MITHICS team.
• Contribute to the drafting of reports, publications and scientific papers arising from the project.
• Contribute to the scientific and technical supervision of staff involved in the project.
Your Profil
Skills
• PhD in cell biology, molecular biology, mitochondrial biology, genetics or a related field.
• Expertise in synthetic biology and approaches to genetics and molecular biology.
• Expertise in the cultivation, transformation and maintenance of S. cerevisiae and E. coli models.
• Expertise in the construction of expression vectors and the validation of genetic constructs.
• Experience in mitochondrial biology, mitochondrial genetics or the study of mitochondrial DNA is highly desirable.
• Good command of scientific English, both written and spoken. Level C1 of the Common European Framework of Reference for Languages (CEFR).
• Proficiency in PCR, RT-qPCR/qPCR and standard molecular biology techniques.
• Proficiency in vector assembly techniques such as Gibson, Golden Gate, Gateway, etc.
• Ability to develop and optimise experimental protocols.
• Proficiency in analysing sequencing data or the ability to liaise with relevant specialists.
• Proficiency in data analysis and processing tools (GraphPad Prism, Excel, ImageJ/Fiji or equivalents).
• Ability to critically analyse results and propose experimental adjustments.
• Scientific independence, rigour, organisational skills and the ability to take the initiative.
• Ability to work as part of a team and in an international collaborative environment.
• Good scientific communication skills and the ability to present results clearly.
• Ability to meet project deadlines and objectives.
Your Work Environment
This recruitment is part of the international MIDAS (Mitochondrial Integration and DNA Access Systems) project, funded by the ARIA programme. This project aims to develop new approaches to mitochondrial genome engineering, ranging from the production and delivery of synthetic mitochondrial DNA to its expression and maintenance within mitochondria. The successful candidate will be involved specifically in WP1, which focuses on the development and validation of new techniques for the synthesis of human mitochondrial DNA. This work will, in particular, enable the study of the impact of mitochondrial variants and haplogroups in various cellular models. The project will be carried out within the MITHICS team at the IBGC (UMR5095, CNRS/University of Bordeaux), which brings to the consortium its expertise in mitochondrial physiology and bioenergetics, pharmacological screening and cellular models, as well as its high-throughput phenotyping approaches. The successful candidate will work in close collaboration with the international partners of the MIDAS consortium, which brings together complementary expertise in mitochondrial biology, synthetic biology, nanocomplex engineering and gene therapy.
