Job Description
Missions
To perform research in molecular and cellular biology, in particular to investigate the regulation of DNA replication using the fission yeast S. pombe as a model organism, with the goal of completing a research article.
Activity
- Plan, carry out, and analyze experiments related to the regulation of DNA replication, in particular the link with cell cycle dynamics. Perform genome-wide analyses of replication origin usage and replication factor binding in different experimental conditions and mutants.
- Discuss and present findings at lab meetings, research seminars, and conferences
- Participate in the overall maintenance and functioning of the lab
- Prepare and write scientific articles that will be submitted for publication in peer-reviewed journals
Your Profil
Skills
- Mastery of classical molecular biology techniques (plasmid construction, western blots, PCR, RNA manipulation...) as well as with genetic engineering approaches (CRISPR)
- Mastery of standard techniques in genetics and cellular biology related to the use of fission yeast as a model system.
- Experience with next-generation sequencing methodologies (ChIP-seq, whole-genome sequencing), including with library preparation protocols for these methods and the associated bioinformatic analysis pipelines.
- Ability to work in a team and in collaboration with internal and external team members and collaborators.
- Excellent level of spoken and written English. The language of the teams is English, both for the interactions between the members of the group and for the writing of research articles.
Your Work Environment
The faithful duplication and transfer of the genetic material is achieved by a tight coordination of DNA replication with cell cycle events, along with multiple pathways that promote genome integrity. This gives rise to a spatial and temporal organization of DNA replication, referred to as the replication program (RP). However, despite our in-depth knowledge of the molecular details of DNA synthesis, key aspects of the mechanisms that establish the RP and the importance of this organization for the cell remain poorly understood. The research project combines a multidisciplinary strategy with a unique model that we have developed in fission yeast to investigate the mechanisms by which cell cycle dynamics shapes the RP. Our work takes advantage of state-of-the-art methodologies, from genome-wide to single-cell, to study the intricate interplay between cell cycle regulation, the organization of DNA replication, and chromatin structure.
The recruited person will work in the Institute of Biochemistry and Cellular Genetics (CNRS UMR 5095). The IBGC is a fundamental research institute focused on various themes ranging from cell cycle biology to the study of metabolism. The institute is made up of 13 research teams that use a wide range of model systems. The Research Engineer will join the team of Dr. Pei-Yun Jenny Wu, which uses fission yeast to study fundamental biological mechanisms.
The Research Engineer will have access to all the experimental and computing resources of the teams as well as to the common services of the IBGC and other core facilities of the Bordeaux Campus.
The Research Engineer will be in contact on a daily basis with the members of the teams of Dr. Wu and Dr. Coudreuse as well as with those from others teams of the IBGC floor where she/he will work (Dr. Royou, Dr. McCusker). She/he will also be in contact with the heads of the IBGC core facilities and, when necessary, of the other common services of the University of Bordeaux.
