Missions

-The engineer will be responsible for carrying out intravital microscopy experiments in an infectious environment as part of a scientific project conducted by a European research consortium.

Activities

Le/la candidat-e sera chargé-e de la mise en œuvre de la microscopie intravitale du poumon chez la souris afin d'analyser la localisation, l'activation et la dynamique des cellules immuntaire lors d'une infection par M. tuberculosis. • Préparation in vitro de cellules immunitaires (culture cellulaire) et transfert adoptif chez la souris (injections intraveineuse, intranasale ou intratrachéale) • Préparation et suivi des modèles infectieux murins • Réaliser l'imagerie intravitale du poumon par microscopie multiphotonique en environnement de niveau de confinement 3 (NSB3, préparation de l'animal et acquisition). • Analyser les données à l'aide de logiciels spécialisés en imagerie (ex. Imaris, Image J). • Réaliser l'analyse par cytométrie en flux des échantillons biologiques issus des modèles et analyser les données à l'aide de logiciels spécialisés en cytométrie (ex. FlowJo). The successful candidate will be responsible for implementing intravital lung microscopy in mice to analyze the localization, activation, and dynamics of immune cells during Mycobacterium tuberculosis infection. • In vitro preparation of immune cells (cell culture) and adoptive transfer into mice (intravenous, intranasal, or intratracheal injections) • Establishment and monitoring of murine infectious models • Performance of intravital lung imaging using multiphoton microscopy in a biosafety level 3 (BSL-3) environment, including animal preparation and image acquisition • Analysis of imaging data using specialized software (e.g., Imaris, ImageJ) • Flow cytometry analysis of biological samples derived from experimental models and data analysis using dedicated cytometry software (e.g., FlowJo)

Skills

We are looking for a candidate with skills in several of the following areas: • Animal experimentation (injections, sampling, analyses on isolated tissues) • Intravital microscopy: preparation of live samples (anesthetized animal) • Fluorescence microscopy (image acquisition and visualization) • Proficiency in image analysis and processing tools (Imaris, ImageJ, or others) • Immunology • Cytometry • Good laboratory research practices •

Work Context

The Institute of Pharmacology and Structural Biology (IPBS) has 230 staff members, including around 80 permanent researchers working in 16 research teams and 4 IBiSA-labeled technological platforms (Proteomics, PICT, Anexplo, and TRI-IPBS). IPBS has two multiphoton microscopes, one of which is in an infectious environment (BSL3). We are one of the few centers in Europe to master intravital lung imaging and to be able to perform it under BSL3 confinement. The successful candidate will receive internal training to work in this environment. The recruited engineer will work under the supervision of Dr. E. Lefrançais (IPBS-Toulouse) on a project in collaboration with a consortium of European teams.
