Missions

The BmrA transporter is an archetype of ABC transporter, allowing the efflux across the membrane of many xenobiotics, allowing to reduce their intracellular concentration and thus to protect the cell against chemical attacks. This defense mechanism gives the cell resistance to many drugs. The team has been studying this transporter for many years using a structural enzymology approach, to better understand the link between enzymology in solution and structural studies in solution.

Activities

The postdoc project is divided into 2 parts:
- Obtain time-resolved data of the Inward-facing to Outward-facing transition of the transporter. These data will allow to obtain the kinetic constants of the different steps of the transport mechanism. This study, already advanced, will be completed by obtaining new kinetic data and obtaining the best enzymatic model, as well as finalizing the structural characterization by cryoEM.
- Obtain liposome structural data for BmrA, to extract the structural movements of the protein by 3DVA analysis and the software developed in the phenix.varref team.

Skills

The candidate will hold a PhD, and will have solid knowledge in structural enzymology. Experience in handling membrane proteins is strongly recommended. The candidate will be comfortable with the concepts of enzymology, liposome preparation, and cryoEM observation as well as data processing with Cryosparc or Relion.

Work Context

The laboratory is located within the Molecular Microbiology and Structural Biochemistry unit (MMSB) in the Lyon Gerland Bioscience Campus, in close proximity to the city center and with convenient access to public transportation, including the subway, tram, and cycle paths. We offer a welcoming environment with a friendly and satisfying working atmosphere, excellent facilities, a workplace with all the benefits of public service, as well as training and development opportunities. As the second largest city in France, Lyon is a prominent center for research on membrane proteins. The candidate will join a collaborative and enthusiastic team, with 3 permanent staff, 1 PostDoc and 2 PhD students on related subjects, as well as M2 students. The stopped-flow experiments will be carried out at the CBS in Montpellier in collaboration with Dr. Corinne Lionne. Short data collection missions are to be expected there.
