Missions
The project aims to better understand molecular mechanisms of infection through structural studies of virulence factors in emerging infectious disease. We focus on Bunyaviruses, three of which are listed among the World Health Organization (WHO) top-ten priority pathogens, including the highly deadly Crimean-Congo hemorrhagic fever virus (CCHFV). The project is supported by outstanding funding over a 5-year period (https://www.fondationbs.org/notre-communaute/laureats-et-projets/anja-bockmann). More on the team's research projects at https://mmsb.cnrs.fr/en/team/protein-solid-state-nmr/.
A key objective of the successful candidate will be the implementation of advanced fast MAS solid-state NMR, and also the set-up of DNP approaches. The goal is to study structures of the viral membrane-bound proteins of interest, as well as their interactions with nucleic acids. S/he will produce samples following well-established protocols relying mainly on cell-free synthesis, but also bacterial expression. S/he will examine them using solid state NMR, possibly complemented by other biophysical approaches and solution NMR. S/he will develop specific selective labeling strategies, as introduction of novel NMR isotopes for combinatorial labeling approaches, to open new opportunities for NMR. The PD will work closely with other lab members involved in different aspects of this larger project.
The position is available from January 2026, with later being possible. The position is initially funded for one year, with possible extension for up to four years.
Activités
- NMR measurements (0.7 mm 1H-detected MAS NMR), implementation of advanced approaches
- NMR spectra analysis (CCPNmr) and data interpretation
- Protein preparation using cell-free/bacterial protein synthesis, design of dedicated labeling variants
- Biophysical characterization of the proteins and their interactions
Compétences
The ideal candidate holds a PhD in chemistry, biochemistry or biophysics (or related discipline), and a strong interest in interdisciplinary work on (viral) proteins. S/he will have experience in structural biology techniques, and possibly a track-record in solution- and/or solid-state NMR spectroscopy. Candidates with a strong background in other structural biochemistry approaches are also encouraged to apply.
Contexte de travail
The protein solid-state NMR laboratory is located within the Molecular Microbiology and Structural Biochemistry (MMSB) unit in the Lyon Gerland Bioscience Campus, regrouping major institutions in virology such as the CIRI and the Lyon BSL4 lab. The campus is located in close proximity to the city center and with convenient access to public transportation, including the subway, tram, and cycle paths. The lab has access to the High Field NMR Center of Lyon (CRMN), housing eight NMR spectrometers (300 MHz, 400 MHz, 500 MHz, 600 MHz, 700 MHz, 2 x 800 MHz and 1000 MHz, some equipped for DNP) and fast-MAS probes (2 x 0.7 mm probes, 1.3 mm probe, …). The MMSB is fully equipped with wet lab for protein expression and purification. Researchers also benefit from access to a range of biophysical platforms (ITC, SEC-MALLS, nanoDSF…) at the MMSB and mass spectrometry through the SFR bioscience platform. We offer a welcoming environment with a friendly and satisfying working atmosphere.
