Job Description
Missions
Respiratory Syncytial Virus RSV is a major public threat, particularly dangerous for children and the elderly, with no therapeutic options available for the vast majority of the population. This position aims at uncovering the mechanisms of RSV replication in the infected cell by cryo-electron tomography and single particle cryo-electron microscopy.
Activity
Sample preparation, optimisation, cryo-EM, cryo-CLEM, cryo-FIB milling and cryo-ET, 3D image analysis and, where appropriate, biochemical and biophysical characterisation of the RSV ribonucleoprotein complexes to understand the mechanisms involved.
Your Profil
Skills
- Proficiency in molecular biology and biochemistry (cloning, expression, purification, biochemical characterisation);
- Expertise in insect and mammalian cell expression systems;
- Experience in molecular virology and viral infection would be an advantage;
- Experience in correlative imaging, cryo-FIB milling, cryo-EM and cryo-ET and data acquisition would be an asset;
- Proficiency in using the computational tools necessary for image processing (Linux, computing cluster, image processing software);
- Solid experience in single particle cryo-EM image analysis;
- Experience in cryo-ET image analysis would be a plus;
- Problem-solving skills, willingness to work in an international, multidisciplinary team;
- Initiative, rigor and organisational skills are essential;
- B2 English level minimum (written/oral).
Your Work Environment
Located on the Grenoble Science Park (ESRF/ILL/EMBL/IBS), the Institute of Structural Biology (IBS) is a national and international player in the field of integrated structural biology. The IBS is a research institute, a technical platform, a host site and a scientific training center affiliated to the Université Grenoble-Alpes, the CNRS and the CEA. It currently employs about 320 scientists, including numerous students and post-docs from all over the world, and benefits from state-of-the-art infrastructure and equipment in cellular and molecular biology, biochemistry, biophysics and structural biology. The IBS is one of the founders of the Partnership for Structural Biology (PSB) with prestigious European institutes: the European Synchrotron Radiation Facility (ESRF), the ILL neutron scattering facilities, and the European Molecular Biology Laboratory (EMBL) Grenoble outstation, all located at the same campus, offering a strong international environment. As a joint research unit (CEA-CNRS-UGA), the IBS is composed of 20 research groups, each of which proposes a multi-disciplinary approach, at the frontiers of biology, physics and chemistry. Within the IBS, the candidate will join the https://www.ibs.fr/en/research/assembly-dynamics-and-reactivity/microscopic-imaging-of-complex-assemblies-mica-group-i-gutsche/ group focused on imaging macromolecular complexes and cells with the goal of gaining structural insights that would yield functional understanding of cellular processes, and with a strong interest in nanoscale exploration of the multiplication of the human RSV.
Constraints and risks
Without.
The research will be carried out in either French or English, however it is expected that the non-French speaking candidate will strive to learn the basics of French in order to facilitate communication and integration into the laboratory.
