Job Description

Missions

The candidate will need to develop a hepatic model using liver organ on chip technology housing multi cellular hepatic lineages from human induced from pluripotent stem cells. Then he will need to expose the model to biological compounds selected by the consortium.

Activity

- Establish multicellular induced pluripotent stem cells cultures using hepatocytes derived cells and LSEC derived cells,
- Transfer the protocol into the organ on chip technology,
- Confirm the cellular model functionality and stability
- Perform biological compounds expositions, selected by the consortium, within the organ on chip model,
- Analysis the results of the experiments,
- Perform the experiments including cell differentiation, organ on chip preparation, organ on chip cultures, 3D spheroids and Petri cultures, cellular and culture medium assays,
- Omic data set analysis and post treatment,
- Report and present the research works to colleagues in meetings, seminars,
- Write papers and communications for workshops/congresses,
- Transfer the technology and know-how to other project members.

Your Profil

Skills

- The candidate should have skills in hiPSCs biology and bio engineering,
- The candidate should be independant to perform cell culture, cellular biology and molecular biology related to liver cells,
- Know how in preparation of samples for omic analysis and data post treatment will be appreciated,
- The candidate should master organ on chip technology and tissue culture inside those technologies,
- The candidate should have practiced liver cells culture and have know how on liver toxicity would be a appreciated,
- The candidate should work in a team and shared his know how and skills with other team members,
- As the work will be done in Japan, the candidate should be able to speak english.

Your Work Environment

The work is the frame of LIMMS and will be performed at Sakai/Nishikawa laboratory at the University of Tokyo, at Hongo campus. LIMMS is a CNRS International Research Laboratory (IRL 2820) on MEMS and NEMS (Micro- and Nano-Electro-Mechanical Systems), jointly operated by France (CNRS), and Japan (University of Tokyo). In this frame, the Sakai/Nishikawa laboratory, located on the Hongo campus is collaborating with LIMMS to develop Organ on chip technology related to liver bioengineering and hepatic development, pathalogy and toxicity. The laboratory of Profs Sakai/Nishikawa develops, among other, organ on chip technology for hepatoxicity, liver stem cells based technology for regenerative medicine and disease recaptulation. The collaboration between LIMMS and Sakai/Nishikawa laboratory is supported by the BPI APOLLO . The candidate will join the researchers involved in BPI APOLLO project at LIMMS/Sakai/Nishikawa laboratory. The candidate will be under the direct supervision of Dr Leclerc, and the under the direction of Dr Clement, director of LIMMS, and Prof Sakai , the director of the Organ and Biosystem engineering/Sakai/Nishikawa laboratory. As such the candidate will have to follow LIMMS and Sakai/Nishikawa laboratory rules. Due to the collaboration between France and Japan, missions to France and inside Japan are expected. Finally, we expected to hire a candidate already residing in Japan. The position opening is subject to funding.

Constraints and risks

- Culture medium change will require to come in the laboratory during the week end.
- The laboratory is located in Tokyo, Japan. Japan has a high risk of natural disasters such as earthquakes, typhoons,…
- Risk related to the utilisation of biolgical samples and human cell cultures,
- Risk related to the manipulation of biological and chemical products,
- Risk related to the manipulation of equipment (laser confocal miscroscope, centrifuges, N2 storage, water temperature bath, …),
- Risk related to ergonomy (desk and computer works).

Compensation

Between €3070 and €3505 gross per month, depending on experience (+ allowances).
