Missions

The postdoctoral researcher's mission will be to study lipid remodeling triggered by phosphate deficiency at the tissue and subcellular levels in plants and algae using Tof-SIMS.

Activities

Preparation of plant or algae samples to establish a new cryofixation and cryosectioning method
- Analysis of samples using Tof-SIMS and data processing
- Interpretation and presentation of results with project partners

Skills

 Knowledge
PhD in biochemistry and/or chemistry; knowledge of lipids is a plus; basic knowledge of mathematics, physics, and data analysis.
Use of mass spectrometry.
Cryomanipulation and cryofixation.
Scientific English (level B1 to B2).

Work Context

The postdoctoral researcher will be part of the MicroMass project of the PEPR BBEST, which brings together four partners (École Polytechnique in Paris, and the LPCV, MEM, and LETI laboratories of the CEA Grenoble). The postdoctoral researcher will work both at the LPCV laboratory, where the biological samples will be produced, and at LETI, where the Tof-SIMS is located.
The aim of the project is, on one hand, to establish a robust data acquisition process, particularly for frozen-hydrated samples (surface preparation, cryo-transfer, study of acquisition parameters) and to correlate these data with those obtained by (cryo-)electron microscopy. The other aim is to interpret the mass spectra, in particular through the synergistic use of MALDI-FT-ICR and ToF-SIMS, in order to evaluate the limits of the technique in terms of qualitative and quantitative detection of different molecules (lipids, carbohydrates).

The position is located in a sector under the protection of scientific and technical potential (PPST), and therefore requires, in accordance with the regulations, that your arrival is authorized by the competent authority of the MESR.
