Description of the thesis topic
Xcc (Xanthomonas campestris pv. campestris) is the causal agent of black rot disease on Brassicaceae (Arabidopsis, cabbage …). A recent study performed in our group (Infectious Strategies of Xanthomonas, LIPM) identified genes important for Xcc adaptation during plant infection using a transposon sequencing (TnSeq) screen. The PhD project will include bacterial fitness analyses using RB-TnSeq, transcriptomic analyses by RNAseq and functional characterization of genes in several plant compartments (leaf surface, hydathodes, mesophyll,…) and ex-planta conditions (rich and minimal media, xylem sap). This work will capture a snapshot of the physiology of Xcc during plant infection and lead to (i) characterization of genes functionally important for bacterial fitness in planta; (ii) characterization of transcriptional regulators important for Xcc transcriptional reprogramming throughout the infectious cycle; (iii) identification of pathways important for Xcc adaptation to distinct plant environments.
Work Context
The Laboratory of Plants-Microbes-environment Interactions (LIPM), UMR2594/441 is part of the National Research Institute for Agriculture, Food and Environment. The lab is located in Toulouse area in the south-west of France.
