The candidate's mission will be to develop and optimize the bioproduction of therapeutic proteins by cell-free systems. He/she is recruited in the framework of a scientific project of the national challenge "Biomedicine".
He/she will be in charge of setting up and optimizing an original cell-free system for the efficient and low-cost production of antimicrobial peptides. This transcription and translation in vitro system will allow the incorporation of non-canonical amino acids into the targeted antimicrobial peptides. The incorporation will use genetic code reprogramming and the recent and versatile technology of flexizymes for tRNA acylation. The candidate will also set up a high-throughput functional assay to evaluate the antimicrobial activity of the peptides produced.
- strong organizational skills, with a strong interest in multidisciplinary and collaborative projects;
- autonomous work and ability to communicate;
- solid knowledge in molecular biology; experience in handling RNA molecules would be a plus;
- skills in recombinant protein expression in Escherichia coli and purification of peptides/proteins by various chromatographic methods;
- oral and written communication skills in both French and English.
The host team “Enzymology and non-ribosomal Peptide Biosynthesis” (https://www.i2bc.paris-saclay.fr/enzymology-and-non-ribosomal-peptide-biosynthesis/) is located at CEA Saclay (20 km southwest of Paris) and is part of the Institute of Integrative Biology of the Cell (I2BC, UMR 9198 CNRS, CEA, Paris Saclay University) (https://www.i2bc.paris-saclay.fr/).
The team is composed of 3 researchers and 1 senior technician. It regularly welcomes M2 students, PhD students and post-docs. The team works on the biological synthesis of natural products and uses synthetic biology approaches to produce improved derivatives of these natural products.
The candidate will be supervised by a researcher and will benefit from technical expertise in protein biochemistry of the technician and an engineer CDD. He will also benefit from molecular biology skills of the other members of the team. He will be required to make short stays in one of the partner laboratories (Synthelis biotech).
