The engineer will be in charge of interactomic methods to study interactions between assembly factors and proteins of the SRP. He/She will prepare the samples for proteomic analysis and will do the affinity purification on the extracts. Cells used will be « knock In » clones (expressing tagged-proteins) or « Knock Out » clones (degradation of an assembly factor, for example) that the engineer will have to prepare by CRISPR/Cas9 editing method. He/She will also validate the interactions found by proteomic using LUMIER IP and Yeast Two-Hybrid tests.


- Cell culture, production of clone by CRISPR/Cas9 method
- Molecular biology : Cloning for CRISPR/Cas9 constructs, Gateway cloning, Y-2H.
- Biochemistry approaches : Affinity purification, sample preparation for Proteomic, LUMIER IP


- A 6-month experience in cell culture and molecular cloning is required
- Experience in “omic” method will be appreciated (interactomic, proteomic, transcriptomic…)

Work Context

The IE will be recruited in the team « Cell biology of RNA » headed by Edouard Bertrand at Institut of Human Genetics (IGH). IGH is composed of 24 teams involved in basic and translational research in Biology and Genetics. The host team is interested in RNP (ribonucleoprotein) biogenesis, especially those including non-coding RNA such as snoRNA, snRNA and 7SL (RNA component of SRP for « signal recognition particle »). Its previous work led to discover several assembly factors including chaperones that are involved in formation of newly synthesized RNP. The CDD will be first for 12 monthes and could be reconducted.
