Missions

The candidate will develop a scientific project focused on the determination of active and inactive conformations of the vasopressin V2 receptor using cryo-electron microscopy in order to understand its mechanism of action. In collaboration with the team of Patrick Bron at the Centre de Biologie Structurale, this project will be performed using molecular pharmacology, protein biochemistry, cryo-electron microscopy, image analysis and reconstruction of the strucutres of the signaling complexes of this physiologically and clinically very important receptor.

Activities

-molecular biology and protein engineering
-determination of pharmacological and functional properties of the different receptor constructs
-expression and purification of membrane receptors and of their canonical signaling partners
-reconstitution of the signaling complexes
-preparation of cryo-electron microscopy samples
-characterization of signaling complexes in vitro
-cryo-electron microscopy and negative stain image analysis
-reconstruction of the structures
-result communication

Skills

-expertise in molecular biology and protein engineering
-experience in cell culture, infection of insect cells and transfection of mammalian cells
-expertise in expression, solubilization and purification of membrane receptors
-expertise in molecular pharmacology, ligand binding and functional assays using fluorescence-based approaches
-preparation of samples for cryo-electron microscopy
-expertise in cryo-electron microscopy and image analysis
-experience in using softwares dedicated to structure reconstruction
-skills in result presentation, both in english and french
-team spirit, autonomy, curiosity

Work Context

The Institute of Functional Genomics (IGF) is a multidisciplinary research centre (CNRS-INSERM-University of Montpellier) which is developing a project focused on the functional genomics of physiological and pathological cellular communications in the fields of neurobiology, endocrinology, oncology and cardiology. There are about 250 people at IGF, with researchers, professors, engineers, technicians, postdoc and student fellows, working in 21 different research teams, 5 technological platforms and common services. The candidate will work both in the "Pharmacology and Structural Biology of Membrane Receptors" team at IGF and in the "Multi-scale Structural Biology" team at Centre of Structural Biology in Arnaud de Villeneuve campus.
